Abstract

Antibiotic resistance is a growing global crisis, rendering once-effective treatments powerless

against infections. The misuse and overuse of antibiotics has accelerated the emergence of resistant pathogens, leading to higher mortality rates and increased healthcare costs. Preliminary findings suggest that the natural product aogacillin enhances the efficacy of existing antibiotics by preventing the development of resistance mechanisms. It is hypothesized that aogacillin combats resistance by blocking an acylation process that drives bacterial resistance mechanisms. To enable this bioactivity profiling, synthesis of aogacillin and structurally related analogs is conducted. In particular, structural features influencing 3-dimensional conformation provides information regarding significant active site interactions. Robust synthesis of these analogs and their mechanisms can pave the way for groundbreaking strategies to curb resistance and restore the effectiveness of vital drugs.
